I’m looking for a Windows equivalent of lsof. Preferably something that’s either an RB plugin or that uses standard shell commands (i.e. no external dependencies). This is so I can build an installer-type program, so the end game is that if the install script says to overwrite a file that's locked, it can display a message asking the user to quit the program.
Handle.exe has been suggested to me. However, even when I run that from an admin account, it complains that it isn't running as an administrator. I can't seem to get runas to fix this for me.

You have changed the property of Handle.exe to "run as administrator"? Just because you are in an admin account does not mean that the app is allowed to run as admin. So right-click on it and check.
